UV-Visible Spectrophotometer: UV-Visible Spectrophotometer is a type of absorption spectroscopy in which light of the ultra-violet region (200-1100 nm.) is absorbed by the molecule. Absorption of the ultra-violet radiations results in the excitation of the electrons from the ground state to higher energy state. The energy of the ultra-violet radiation that is absorbed is equal to the energy difference between the ground state and higher energy states. UV-Visible is spectroscopy can be used to determine the concentration of the absorber in a solution. It is necessary to know how quickly the absorbance changes with concentration. This can be taken from references or more accurately, determined from a calibration curve. |
